Key elements and design rationale

  • Target: SCARB1 (Scavenger receptor class B member 1).
  • Antibody format: Monoclonal; clone CGE-19; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

SCARB1 (protein: Glycogen synthase kinase-3 beta (gsk3b)) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Receptor for different ligands such as phospholipids, cholesterol ester, lipoproteins, phosphatidylserine and apoptotic cells. Facilitates the flux of free and esterified cholesterol between the cell surface and extracellular donors and acceptors, such as high density lipoproteins (HDL) and to a lesser extent, apoB-containing lipoproteins and modified lipoproteins (By similarity). Necessary for selective HDL-cholesterol uptake (PubMed:26965621). Probably involved in the phagocytosis of apoptotic cells, via its phosphatidylserine binding activity. Receptor for hepatitis C virus glycoprotein E2. Binding between SCARB1 and E2 was found to be independent of the genotype of the viral isolate (By similarity). . Reported cellular localization context: Cell membrane ; Multi-pass membrane protein. Membrane, caveola; Multi-pass membrane protein. Predominantly localized to cholesterol and sphingomyelin-enriched domains within the plasma membrane, called caveolae. Tissue expression notes (as provided): Widely expressed.

Research relevance and current trends

  • Research context keywords from the source record include: Cancer,Cancer Metabolism,Cardiovascular,Cholesterol Metabolism,Host-Virus Interaction,Interspecies Interaction,Lipid and Lipoprotein Metabolism,Lipid Metabolism,Lipids/Lipoproteins,Metabolic Signaling Pathway,Metabolic Signaling Pathways,Metabolism,Metabolism of Lipids and Lipoproteins,Microbiology,Pathways and Processes,Signal Transduction.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.

Workflow ideas (metafield): Validate SCARB1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect SCARB1 expression by Western blot in cell or tissue lysates, Detect SCARB1 in FFPE tissue sections by immunohistochemistry, Localize SCARB1 by immunofluorescence/immunocytochemistry in cultured cells, Quantify SCARB1-positive cells by flow cytometry in single-cell suspensions

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 110 kDa; calculated MW: 60878 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 110 kDa
  • Cellular localization (provided): Cell membrane ; Multi-pass membrane protein. Membrane, caveola; Multi-pass membrane protein. Predominantly localized to cholesterol and sphingomyelin-enriched domains within the plasma membrane, called caveolae.
  • Tissue details (provided): Widely expressed.

Does M01093 anti-SCARB1/Sr Bi Rabbit Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
You can see on the product datasheet, M01093 anti-SCARB1/Sr Bi Rabbit Monoclonal antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
