Overview
Anti-SLIT1 Antibody is an antibody targeting Slit Guidance Ligand 1, Slit Homolog 1 Protein, Multiple EGF-Like Domains Protein 4, MEGF4 Polyclonal raised in Rabbit (Unconjugated). This antibody is commonly used in IHC, WB to detect, localize, or compare expression of the target across samples.
Key elements and design rationale
- Target: Slit Guidance Ligand 1, Slit Homolog 1 Protein, Multiple EGF-Like Domains Protein 4, MEGF4 (also reported as Slit Guidance Ligand 1, Slit Homolog 1 Protein, Multiple EGF-Like Domains Protein 4, MEGF4).
- Immunogen/epitope region: Secreted.
- Homology note: Human, rat - identical (informative for cross-species interpretation).
- Species reactivity (as provided): Human, Rat, Mouse.
- Specificity statement (as provided): This antibody is specific to SLIT1, it won’t recognize the highly homologous protein SLIT2..
- Lot quality control (as provided): Western blot analysis.
- Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
- Blocking peptide: Available for antigen preadsorption control where appropriate.
These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.
Biological background
Slit homolog 1 protein (SLIT1), also known as multiple EGF-like domains protein 4, and MEGF4, is a secreted protein member of the SLIT family involved in axon guidance and cell migration.SLIT1 is a member of the SLIT family proteins that includes three members: SLIT1, SLIT2 and SLIT3. SLITS are secreted glycoproteins comprised of four stretches of leucine-rich repeat (LRR) domains, nine epidermal growth factor (EGF) repeats, a Laminin-G-like domain, and a C-terminal cysteine knot.1,2SLITS exert their many physiological roles by binding to Roundabout (Robo) receptors family, that includes four members (Robo 1 to Robo 4), which are single transmembrane receptors belonging to the immunoglobulin superfamily.1,2SLITS play a crucial role in axon guidance during the development of the nervous system. Axon guidance is the process by which neurons extend their axons to reach their appropriate targets, such as other neurons, muscles, or glands.
Research relevance and current trends
- Comparing target expression across perturbations, genotypes, or treatment conditions.
- Interpreting localization shifts alongside pathway or phenotypic readouts.
- Using orthogonal controls (KO/KD, peptide competition, isotype concepts) to support conclusions.
Common research applications
- Western blot (WB): compare target abundance/size across lysates and conditions; consider isoforms/PTMs.
- Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.
Notes for experimental interpretation
- Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
- Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
- Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
- Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
- Provided control suggestions: Negative control: BLP-NR221.
- Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
