Biological background

Protein function (datasheet): Receptor-regulated SMAD (R-SMAD) that is an intracellular signal transducer and transcriptional modulator activated by TGF-beta (transforming growth factor) and activin type 1 receptor kinases. Binds the TRE element in the promoter region of many genes that are regulated by TGF-beta and, on formation of the SMAD2/SMAD4 complex, activates transcription. May act as a tumor suppressor in colorectal carcinoma. Positively regulates PDPK1 kinase activity by stimulating its dissociation from the 14-3-3 protein YWHAQ which acts as a negative regulator. .

Cellular localization (datasheet): Cytoplasm. Nucleus. Cytoplasmic and nuclear in the absence of TGF-beta. On TGF-beta stimulation, migrates to the nucleus when complexed with SMAD4. On dephosphorylation by phosphatase PPM1A, released from the SMAD2/SMAD4 complex, and exported out of the nucleus by interaction with RANBP1.

Tissue details (datasheet): Expressed at high levels in skeletal muscle, endothelial cells, heart and placenta. .

Research relevance and current trends

  • Commonly studied in contexts related to Cancer,Cytoplasmic,Epigenetics and Nuclear Signaling,Growth Factors,Nuclear Signaling,Nuclear Signaling Pathways,Signal Transduction,Signaling Pathway,Stem Cells,TGF Beta.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

We are currently using anti-Smad2 Rabbit Monoclonal antibody M00090 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on feline tissues as well?
The anti-Smad2 Rabbit Monoclonal antibody (M00090) has not been tested for cross reactivity specifically with feline tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in feline you can get your next antibody for free. Please contact me if I can help you with anything.
We were well pleased with the WB result of your anti-Smad2 Rabbit Monoclonal antibody. However we have been able to see positive staining in colon adenocarcinoma cytoplasm using this antibody. Is that expected? Could you tell me where is SMAD2 supposed to be expressed?
From literature, colon adenocarcinoma does express SMAD2. Generally SMAD2 expresses in cytoplasm. Regarding which tissues have SMAD2 expression, here are a few articles citing expression in various tissues: Cervix carcinoma, Pubmed ID: 18669648, 18691976, 20068231 Cervix carcinoma, and Erythroleukemia, Pubmed ID: 23186163 Chronic myeloid leukemia cell, Pubmed ID: 8980228 Colon adenocarcinoma, Pubmed ID: 9702198 Kidney, Pubmed ID: 8752209 Kidney, Pancreas, and Spleen, Pubmed ID: 15489334 Placenta, Pubmed ID: 8774881
