Product Overview
This rabbit polyclonal antibody detects suppressor of fused homolog (gene SUFU) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 53.9 kDa, and the supplier reports an observed band at about 54 kDa.
Suppressor of fused (SUFU) is a key negative regulator of Hedgehog signaling. It binds the GLI transcription factors and holds them in the cytoplasm, and it promotes processing of GLI3 into its repressor form; Hedgehog pathway activation releases the GLI proteins to enter the nucleus. SUFU mutations predispose to medulloblastoma and Gorlin syndrome.

Samples with a confirmed band (WB): human HEL cells, human 293T cells, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): U937 cells.

Recombinant human SUFU fragment (Leu34–His484), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places suppressor of fused homolog in the cytoplasm, nucleus and cell projection. Tissue expression noted by UniProt (human): Ubiquitous in adult tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
