Overview
Guinea pig Anti-EMR1 (ADGRE1) (extracellular) Antibody is an antibody targeting MR1 hormone receptor, EGF-like module-containing mucin-like hormone receptor-like 1, Adhesion G protein-coupled receptor E1, TM7LN3, F4/80, GPF480 Polyclonal raised in Guinea Pig (Unconjugated). This antibody is commonly used in WB to detect, localize, or compare expression of the target across samples.
Key elements and design rationale
- Target: MR1 hormone receptor, EGF-like module-containing mucin-like hormone receptor-like 1, Adhesion G protein-coupled receptor E1, TM7LN3, F4/80, GPF480 (also reported as MR1 hormone receptor, EGF-like module-containing mucin-like hormone receptor-like 1, Adhesion G protein-coupled receptor E1, TM7LN3, F4/80, GPF480).
- Immunogen/epitope region: Extracellular, N-terminus.
- Homology note: Mouse, Rat - 11/14 amino acid residues identical (informative for cross-species interpretation).
- Species reactivity (as provided): Human, Rat, Mouse.
- Lot quality control (as provided): Western blot analysis.
- Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
- Blocking peptide: Available for antigen preadsorption control where appropriate.
- Conjugate/format: Unconjugated (may affect detection channel and background).
These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.
Biological background
Human epidermal growth factor (EGF)-like module containing mucin-like hormone receptor 1 (EMR1) is a surface receptor with unknown function that belongs to the adhesion family of the G protein-coupled receptors (GPCR). This receptor is encoded by the ADGRE1 and like all GPCRs, contains seven transmembrane domains, an intracellular C-terminus and an extracellular N-terminus. The NH2 domain has six EGF-like domains.Adhesion GPCRs share a long N-terminal extracellular sequences comprising multiple domains, separated from the transmembrane segments by a serine/threonine-rich domain, a feature reminiscent of mucin-like, single-span, integral membrane glycoproteins with adhesive properties1,2.EMR1 protein is a homolog of F4/80, a murine protein that is widely being used as a marker of murine macrophage populations1.Human EMR1 is exclusively expressed on mature eosinophils in the blood and bone marrow and in nasal polyps.
Research relevance and current trends
- Comparing target expression across perturbations, genotypes, or treatment conditions.
- Interpreting localization shifts alongside pathway or phenotypic readouts.
- Using orthogonal controls (KO/KD, peptide competition, isotype concepts) to support conclusions.
Common research applications
- Western blot (WB): compare target abundance/size across lysates and conditions; consider isoforms/PTMs.
Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.
Notes for experimental interpretation
- Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
- Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
- Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
- Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
- Provided control suggestions: Negative control: BLP-ER051.
- Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
