Overview

HLA-B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exon 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. Hundreds of HLA-B alleles have been described.

This anti-HLA-B antibody is supplied as Purified (Mouse, Monoclonal (mouse origin), clone SPM420, Mouse IgG, kappa, Unconjugated) and is designed to support common target-detection workflows after the on-page specifications.

Key elements and design rationale

  • Target: HLA-B
  • Format: Purified
  • Localization: Cell Surface
  • Species reactivity: Human
  • Applications (listed): FACS, IF
  • Conjugate: Unconjugated
  • Clone and antibody class: Monoclonal (mouse origin), clone SPM420, Mouse IgG, kappa

Because antibody performance can depend on epitope context, sample preparation, and biological state, interpret signals using appropriate controls and orthogonal evidence when possible.

Biological background

HLA-B is referenced in public gene/protein resources (e.g., UniProt and NCBI Gene), which provide curated names/synonyms, protein features, and pathway context. When designing assays, consider potential isoforms, post-translational modifications, and cell-type specific expression that may influence observed signal.

Research relevance and current trends

  • Profiling HLA-B expression across model systems, perturbations, and time points to support mechanistic hypotheses.
  • Combining antibody-based detection with multi-omics or imaging readouts to link HLA-B signal with phenotype.
  • Using well-matched controls (isotype controls, genetic perturbations, or independent reagents) to strengthen interpretation of target-associated signal.

Common research applications

  • FACS
  • IF

Use the listed applications as a starting point and tailor experimental design to your sample type and readout requirements.

Notes for experimental interpretation

  • Specificity considerations: closely related family members, isoforms, or PTMs can affect apparent specificity; confirm with independent approaches when critical.
  • Controls: include negative controls and, when feasible, genetic or pharmacologic perturbations to support target attribution in your system.
  • Species and sample context: differences in sequence, expression, fixation, or extraction conditions can change signal behavior across models.
