Isotype controls are primary antibodies that lack specificity to the target, but match the class and type of the primary antibody used in the application. Isotype controls are used as negative controls to help differentiate non-specific background signal from specific antibody signal. Our Rabbit IgG Isotype Control-ATTO Fluor-550 (#RIC-001-AO), was specifically developed to be used as a negative control in immunohistochemistry and surface staining flow cytometry applications, along our line of rabbit primary antibodies conjugated to the ATTO Fluor-550 fluorophore.
Host Rabbit
Conjugate(s) ATTO-550. Maximum absorption 554 nm; maximum fluorescence 576 nm. The fluorescence is excited most efficiently in the 540 - 565 nm range. This label is related to the dye Rhodamine 6G and can be used with filters used to detect Rhodamine.
Application(s) FC, IHC

Overview

Rabbit IgG Isotype Control-ATTO Fluor-550 is an antibody Polyclonal raised in Rabbit (ATTO-550. Maximum absorption 554 nm; maximum fluorescence 576 nm. The fluorescence is excited most efficiently in the 540 - 565 nm range. This label is related to the dye Rhodamine 6G and can be used with filters used to detect Rhodamine.). This antibody is commonly used in FC, IHC to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Specificity statement (as provided): The IgG fraction was purified from non-immunized rabbits..
  • Lot quality control (as provided): Flow cytometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: ATTO-550. Maximum absorption 554 nm; maximum fluorescence 576 nm. The fluorescence is excited most efficiently in the 540 - 565 nm range. This label is related to the dye Rhodamine 6G and can be used with filters used to detect Rhodamine. (may affect detection channel and background).
  • Antibody type: Polyclonal / Rabbit IgG.

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

This target is commonly studied across diverse experimental systems; interpretation benefits from considering cell type, isoforms, and stimulation state.

Research relevance and current trends

  • Comparing target expression across perturbations, genotypes, or treatment conditions.
  • Interpreting localization shifts alongside pathway or phenotypic readouts.
  • Using orthogonal controls (KO/KD, peptide competition, isotype concepts) to support conclusions.

Common research applications

  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
