- Karyotype: The stemline chromosome number is hypotriploid with the modal number of 56 chromosomes per cell and the 2S component occurring at 13.2%. Over two-thirds of the chromosome complement consisted of structurally rearranged chromosomes. Most marker chromosomes had complex rearrangements. The origin of the segments composing these markers could not be identified. Of the identifiable markers, 6p+/q+, 7p+, 11p+, and 12p+ occasionally were present at 2 copies per cell. The Y chromosome was not detected in the QM stained preparation.

- subculturing: Remove the old medium from the adherent cells and wash them with PBS that lacks calcium and magnesium. For T25 flasks, use 3-5 ml of PBS, and for T75 flasks, use 5-10 ml. Then, cover the cells completely with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks. Let the cells incubate at room temperature for 8-10 minutes to detach them. After incubation, gently mix the cells with 10 ml of medium to resuspend them, then centrifuge at 300xg for 3 minutes. Discard the supernatant, resuspend the cells in fresh medium, and transfer them into new flasks that already contain fresh medium.

SaOS-2 (Sarcoma OSteogenic-2) is a human osteosarcoma cell line with adherent, epithelial-like morphology, derived from a young female patient. Cellosaurus: CVCL_0548.
Culture SaOS-2 in DMEM:Ham's F12 (1:1) with 10% FBS. The cells are adherent; dissociate with Accutase and seed at approximately 1 x 10^4 cells/cm^2 (doubling time about 35-40 hours).
Adherent - SaOS-2 grows as an epithelial-like monolayer.
SaOS-2 is used in bone biology and osteoblast differentiation and mineralization studies, and as an osteosarcoma model.
Supplied as cryopreserved cells and shipped on dry ice. Each batch is quality-controlled by STR profiling (identity) and mycoplasma testing, and comes with a batch-specific Certificate of Analysis (CoA).
