1-Stearoyl-2-linoleoyl-sn-glycero-3-phosphocholine, also known as SLPC and 18:0-18:2 PC, is an endogenous phospholipid marker in the glycerophospholipid metabolic pathway and a core component of the phospholipid bilayer of cell membranes and high-density lipoprotein (HDL), where it helps regulate lipoprotein functional homeostasis and can serve as a key lipid responsive to radiation injury and cardiometabolic disease. Its level decreases in a dose-dependent manner in mouse serum as ionizing radiation dose increases and decreases significantly in human HDL in metabolic syndrome, so it can serve as a biological dosimeter for rapid, accurate assessment of radiation absorbed dose in exposed individuals and as a lipidomics research target for cardiometabolic diseases such as lipid metabolic syndrome and early-onset coronary heart disease[1][2]. In Vitro
In mouse serum lipidomics LC-MS assays, 1-Stearoyl-2-linoleoyl-sn-glycero-3-phosphocholine (endogenous relative quantitative concentration in mouse serum, measured by ex vivo serum treatment 6 h after ionizing radiation exposure) is identified as an ionizing radiation-responsive lipid. Its serum abundance falls significantly as 60Co source γ-ray doses increase across 0-8 Gy, and it correlates strongly and negatively, in a linear manner, with radiation dose[2]. In mouse serum lipidomics LC-MS assays, 1-Stearoyl-2-linoleoyl-sn-glycero-3-phosphocholine (endogenous relative quantitative concentration in mouse serum, measured by ex vivo serum treatment 72 h following ionizing radiation exposure) displays a significant dose-dependent decrease in serum abundance across 60Co source γ-ray irradiation doses of 0-8 Gy, together with a strong linear negative correlation with radiation dose[2]. In human HDL lipidomics LC-MS detection experiments, the level of 1-Stearoyl-2-linoleoyl-sn-glycero-3-phosphocholine in the HDL3 subfraction is 15% lower in subjects with metabolic syndrome than in non-diseased subjects[2].
