5-NIdR (1-(β-D-2-Deoxyribofuranosyl)-5-nitroindole) is an artificial nucleoside capable of inhibiting replication of the DNA lesions generated by Temozolomide. It induces apoptosis in cancer cells and arrests the cell cycle at the G0 phase, and it enhances the anti-tumor efficacy of Temozolomide in a murine glioblastoma model[1]. It is supplied as a light yellow to yellow solid (C13H14N2O5, MW 278.26) at 99.81% purity.

In Vivo

Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 400 μL PEG300; then 50 μL Tween-80; then 450 μL saline to bring the volume to 1 mL. Saline: dissolve 0.9 g sodium chloride in ddH2O and make up to 100 mL.

How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 900 μL 20% SBE-β-CD in saline. 20% SBE-β-CD in saline: dissolve 2 g SBE-β-CD powder in 10 mL saline until clear (4°C, store up to one week).

In Vitro

5-NIdR (12.5-100 μM; 24-72 h) inhibits the growth of the human glioblastoma cell lines U87, A172, and SW1088 in a dose-dependent manner[1]. In U87 cells, 5-NIdR (1-100 μg/mL; 72 h) induces apoptosis, and overnight incubation with 100 μg/mL arrests the cell cycle at G0 phase[1].

In Vivo

In a murine xenograft model of glioblastoma, 5-NIdR (100 mg/kg; ip; for 5 consecutive days) combined with Temozolomide (40 mg/kg) results in complete tumor regression, whereas Temozolomide alone only delayed tumor growth[1].
