5-Octyl-α-ketoglutarate, also known as 5-Octyl 2-oxopentanedioate, is a cell-permeable substrate for lysine demethylase. It is involved in protein demethylation, inhibits proliferation of wtCRBN-expressing cells, and enhances Lenalidomide-induced sensitivity while inhibiting myeloma (MM) drug resistance in cells. In MM cells undergoing glutamine-dependent proliferation, α-ketoglutarate increases glutamine catabolism to promote protein demethylation[1]. It has a molecular formula of C13H22O5 and a molecular weight of 258.31 g/mol.
