Compound Overview
8-Azido-ATP, also known as 8-Azidoadenosine 5'-triphosphate or 8-N3-ATP, is a photoreactive nucleotide analog useful for identifying proteins such as DNA-dependent RNA polymerase[1]. It is a click chemistry reagent bearing an azide group that can undergo copper-catalyzed azide-alkyne cycloaddition (CuAAc) with alkyne-containing molecules, as well as strain-promoted alkyne-azide cycloaddition (SPAAC) with molecules bearing DBCO or BCN groups. It has the molecular formula C10H15N8O13P3 and a molecular weight of 548.19 g/mol.

In Vitro
8-Azido-ATP can serve to identify the viral RNA polymerase, since the analog inhibits transcription activity associated with rotavirus particles after UV light exposure[1]. 8-Azido-ATP (azido-ATP) lowers viral transcription in a dose-dependent manner[1]. 8-Azido-ATP blocks Kir6.2ΔC26 currents less potently than ATP does: its half-maximal inhibition (Ki) is 2.8 ± 0.4 mM (n = 6), against 172 ± 7 mM (n = 6) for ATP. The Hill coefficients were 0.9 ± 0.2 (8-azido-ATP) and 1.3 ± 0.1 (ATP)[2].
