In Vitro
Against MSSA ATCC 25923 and MRSA ATCC 33591, 8-Demethyleucalyptin shows bacteriostatic activity, with MIC values of 31.25 μg/mL and 62.5 μg/mL, respectively[1]. At 15.625-31.25 μg/mL for 48 h, 8-Demethyleucalyptin inhibits biofilm formation of MRSA and MSSA, with inhibition rates of 31.6% and 37.1%[1]. Staphyloxanthin biosynthesis in MRSA and MSSA is weakly inhibited by 8-Demethyleucalyptin (15.625-31.25 μg/mL; 48 h), with inhibition rates of 14.2% and 22.2%, and acid tolerance is reduced[1]. At 31.25-62.5 μg/mL for 18 h, 8-Demethyleucalyptin disrupts the integrity of the cell membrane and wall in MSSA and MRSA[1]. Over 48-72 h, 8-Demethyleucalyptin inhibits the viability of HCT116 and MRC-5 cells (IC50 = 6.4 and 7.0 μM, respectively), while the IC50 for Jurkat cells is >100 μM[2].
