8-Deoxylactucin is an orally active sesquiterpene lactone that inhibits LPS-induced NO production in RAW264.7 macrophages with an IC50 of 4.35 μM and exerts anti-inflammatory effects through blockade of the NF-κB pathway. It also shows hepatoprotective efficacy in a mouse model of LPS/D-galactosamine-induced acute hepatitis, supporting its use in studies of inflammatory disease and inflammatory liver injury[1]. In Vitro
In RAW264.7 macrophages, 8-Deoxylactucin (1 h pretreatment) significantly suppresses LPS-induced NO production, yielding an IC50 of 4.35 μM[1]. At 1-10 μM and 1 h pretreatment, 8-Deoxylactucin significantly lowers iNOS mRNA and protein expression induced by LPS, along with mRNA expression of the pro-inflammatory cytokines IL-1β, IL-6, and TNF-α, in RAW264.7 cells[1]. In RAW264.7 macrophages, 8-Deoxylactucin (1-10 μM, 2 h pretreatment) significantly prevents LPS-induced phosphorylation of IKKα/β and IκBα, inhibits IκBα degradation, and lowers nuclear accumulation of NF-κB p65[1]. In RAW264.7 cells, 8-Deoxylactucin (1-10 μM, 1 h pretreatment) significantly increases protein expression and nuclear translocation of NRF2, up-regulates protein expression of the NRF2-targeted antioxidant genes (HO, NQO1), and lowers ROS production[1].
