Scientific background

A-192621 is a synthetic compound that acts as a selective antagonist of the endothelin B (ET-B) receptor, a member of the G-protein coupled receptor (GPCR) superfamily.In vivo efficacy studies showed that A-192621 inhibits ET-1-induced dilatory and pressor responses that are mediated by ET-B. The compound can cause an increase in arterial blood pressure and plasma ET-1 levels1,2. It inhibits ET-B receptors with an IC50 value of 4.5 nM3.ET-B receptors play an important role in regulating renal function and blood pressure and are expressed in sensory nerves. They are mainly present in medium- and large-sized cell bodies of human trigeminal ganglia4,5.
