Compound Overview
Acetyl Coenzyme A trisodium (Acetyl-CoA trisodium) is a membrane-impermeant central metabolic intermediate that participates in the TCA cycle and oxidative phosphorylation. It regulates various cellular mechanisms by serving as the sole donor of acetyl groups to target amino acid residues in post-translational protein acetylation, and it is also a key precursor of lipid synthesis[1][2][3][4]. It is supplied as a white to off-white solid (C23H35N7Na3O17P3S, MW 875.52) at 99.22% purity.

In Vitro
| Solvent | Solubility | Notes |
|---|---|---|
| H2O | 83.33 mg/mL (95.18 mM) | requires sonication |
Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); sealed storage, away from moisture; avoid repeated freeze-thaw cycles.
If water is used as the stock solvent, dilute to the working solution and sterilize it through a 0.22 μm filter before use.
In Vivo
Choose the formulation that suits the animal model and route of administration. Percentages are volume ratios of the final working solution. Prepare the working solution fresh on the day of dosing; if precipitation or phase separation occurs, gentle warming or sonication can help.
Direct preparation of the working solution
These formulations are prepared directly, without a DMSO stock; use them promptly after preparation.
Protocol 1
| Composition | PBS |
|---|---|
| Result | 100 mg/mL (114.22 mM); clear solution; requires sonication |

In Vitro
Acetyl coenzyme A trisodium raises cytoplasmic protein acetylation and lowers starvation-induced autophagic fluxes in starved U2OS cells (U2OS cells stably expressing GFP-LC3 are microinjected with Acetyl coenzyme A trisodium, incubated under nutrient-free conditions with 100 nM BafA1, and fixed after 3 h)[2].
In Vivo
In a mouse cardiac pressure overload model, Acetyl coenzyme A trisodium blunts pressure overload-induced cardiomyopathy by suppressing maladaptive autophagy[2][3]. In mice deprived of food for 24 h (with water available ad libitum), total Acetyl coenzyme A trisodium levels fall significantly in several organs, including the heart and muscles, matching a decrease in protein acetylation levels. Under the same conditions, however, Acetyl coenzyme A trisodium concentrations in the brain are not majorly affected, while hepatic Acetyl coenzyme A trisodium and protein acetylation levels actually rise[4].
