The Acid Phosphatase Activity Colorimetric Assay Kit applies a colorimetric format to this measurement, read at 405 nm (reference 630 nm). Hands-on and incubation time is less than 30 minutes.

The kit uses p-nitrophenyl phosphate (pNPP) as a phosphatase substrate which turns yellow (λmax = 405 nm) when dephosphorylated by AP.
