Compound Overview

AcrB-IN-4 is an inhibitor of the AcrB efflux pump, able to potentiate the effect of antibiotics. It inhibits efflux of Nile Red, a known AcrB substrate, without disrupting the bacterial outer membrane or showing toxicity in a nematode model[1]. It has a molecular formula of C29H34N2O4S and a molecular weight of 506.66 g/mol.

Safety

For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with your institution's chemical hygiene plan.

In Vitro

At 8-128 μg/mL, Efflux pump-IN-4 displays outstanding synergistic antibacterial activity with at least one of the antibiotics (ERY, LEV, and MIN). Efflux pump-IN-4 is synergistic with MIN, and at 8 μg/mL it lowers the MIC value of LEV by 4-fold[1]. Efflux pump-IN-4 (50 μM, 100 μM) strongly inhibits Nile Red efflux, already at the lowest concentration of 50 μM[1]. Efflux pump-IN-4 (4-256 μg/mL) causes no hemolysis of mouse red blood cells[1].

In Vivo

Efflux pump-IN-4 (128 μg/mL; 72 h) shows no significant in vivo toxicity toward Caenorhabditis elegans[1].
