Compound Overview
AG-7404 is an orally active, irreversible inhibitor of the picornavirus 3C protease. It blocks the processing of viral polyproteins, thereby inhibiting viral replication, and it has synergistic antiviral activity with capsid inhibitors such as V-073 or BTA798 and remains effective against V-073-resistant variants. It is used in the study of enterovirus infections such as poliovirus[1][2][3]. It is supplied as a white to off-white solid (C26H29N5O7, MW 523.54) at 98.45% purity.

In Vitro
In HeLa cells infected with poliovirus, AG-7404 (0.080-0.674 μM; 3 d) suppresses viral replication, with an EC50 of 0.080-0.674 μM[1]. Against V-073-resistant poliovirus strains, AG-7404 (0.218-0.819 μM; 3 d) stays active, giving an EC50 of 0.218-0.819 μM[1].
