In Vitro

Within the tested dose range, Agathic acid shows no activity against A2780 cells[1]. Agathic acid (72 h) inhibits proliferation of Leishmania amazonensis promastigotes and amastigotes, giving IC50 values of 28.0 μg/mL and 17.0 μg/mL, respectively[2]. In in vitro-cultured Leishmania amazonensis amastigotes, Agathic acid (100 μg/mL; 3 h) significantly raises plasma membrane permeability, with 63.4% of cells positive for Propidium Iodide uptake. It also induces mitochondrial membrane potential depolarization, lowering the proportion of cells with Rhodamine 123 fluorescence to 27.5%[2]. In human erythrocytes, Agathic acid (3 h) shows a CC50 of 350 μg/mL, with a selectivity index of 20.6 against Leishmania amazonensis amastigotes[2].

In Vivo

In a healthy, late-gestation mixed-breed beef cattle model, Agathic acid (102-144 mg/kg; oral administration; twice daily until abortion) causes preterm labor and abortion[3].

Detected agathic acid, dihydroagathic acid, and tetrahydroagathic acid in serum samples, with higher serum concentrations of agathic and dihydroagathic acid in cows that aborted earlier.
