Product Overview
The kit extracts high purity total DNA from blood clot samples as well as from animal tissues, cells, blood and bacteria. It is suitable for 100 µL–1 mL blood clot samples and purifies DNA fragments up to 50 kb in length. The purification process does not require toxic solvents such as phenol or chloroform, and ethanol precipitation is not required. Purified DNA can be used directly in downstream experiments such as digestion, PCR, Real-Time PCR, library construction and Southern hybridization.
The blood clot is lysed with 1–2.5 volumes of Solution RCL, then 200 µL of Solution GAS1, 20 µL of Proteinase K and 300 µL of Solution GA2 are added and the sample is incubated at 56°C for 10 min. After adding 300 µL of absolute ethanol the lysate is loaded onto Adsorption column G, washed with 500 µL of Buffer RP and twice with 600 µL of Wash Buffer, dried by centrifugation, and eluted with 50–200 µL of Solution GE after 2–5 min at room temperature. Purified DNA has an OD260/OD280 of 1.7–1.9.

Usage Notes
- Avoid repeated freeze–thaw cycles.
- Dissolve precipitated Solution GAS1 or Solution GA2 at 56°C before use.
- Add RNase A separately if RNA removal is required; it is not supplied in the kit.
- Add ethanol to the buffers before use.
- Remove residual ethanol before elution to avoid inhibition of downstream enzymes.
