Product Overview
This rabbit polyclonal antibody detects tumor necrosis factor ligand superfamily member 9 (gene Tnfsf9) in mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 33.9 kDa, and the supplier reports an observed band at about 27 kDa.
4-1BB ligand (4-1BBL, TNFSF9) is a TNF family cytokine expressed on antigen-presenting cells that binds 4-1BB (TNFRSF9) on T cells. It supports the proliferation and survival of activated T cells and may take part in activation-induced cell death and T-cell interactions with B cells and macrophages, making it relevant to costimulation research.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Mouse) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat brain tissue, rat lung tissue, rat C6 cells, mouse brain tissue, mouse thymus tissue, mouse lung tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): ANA-1 cells.
Immunogen
Recombinant mouse 4-1BBL/Tnfsf9 fragment (Met1–Glu309), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places tumor necrosis factor ligand superfamily member 9 in the membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
