Product Overview

This rabbit polyclonal antibody detects 5-hydroxytryptamine receptor 2B (gene HTR2B) in human and rat samples and is validated for Western blot. The predicted molecular weight is 54.3 kDa, and the supplier reports an observed band at about 54 kDa.

The serotonin 2B receptor (5-HT2B, HTR2B) is a Gq/G11-coupled G protein-coupled receptor that activates phospholipase C signaling. It is expressed in the heart, gut and other peripheral tissues as well as in the brain, and it is studied in cardiac development, heart valve biology and the side effects of serotonergic drugs.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Rat)

Samples with a confirmed band (WB): rat brain tissue, U87 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide from the C-terminal region of human 5HT2B Receptor. It differs from the mouse sequence at six residues and from the rat sequence at nine residues.

Reactivity Notes

The supplier lists reactivity with human and rat. UniProt places 5-hydroxytryptamine receptor 2B in the cell membrane and synapse. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 54.3 kDa; observed band at about 54 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human 5HT2B Receptor. It differs from the mouse sequence at six residues and from the rat sequence at nine residues.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
