Product Overview
This rabbit polyclonal antibody recognizes ATP-binding cassette sub-family G member 8 (gene ABCG8) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 75.7 kDa, and the supplier reports an observed band at about 76 kDa.
ABCG8 pairs with ABCG5 to form a sterol transporter in the intestine and liver that returns cholesterol and plant sterols to the gut lumen and secretes them into bile. The heterodimer is essential for sterol balance, and ABCG8 variants are linked to sitosterolemia and gallstones.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human, Rat) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human A431 cells, rat small intestine tissue, mouse liver tissue, rat RH35 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (ICC/IF): HepG2 cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells, RH35 cells.
Immunogen
Recombinant human ABCG8 fragment (Arg50–Asp672), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ATP-binding cassette sub-family G member 8 in the cell membrane and apical cell membrane. Tissue expression noted by UniProt (human): Predominantly expressed in the liver. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
