Product Overview

Clone AAOG-1 is a rabbit monoclonal antibody that recognizes actin, alpha skeletal muscle (gene ACTA1) in human, mouse and rat samples and is validated for Western blot, ICC/IF and flow cytometry. The predicted molecular weight is 42.1 kDa, and the supplier reports an observed band at about 42 kDa.

Skeletal muscle alpha-actin (ACTA1) is the main actin isoform in the thin filaments of adult skeletal muscle. It interacts with myosin to generate contraction, and mutations in ACTA1 cause congenital myopathies such as nemaline myopathy.

Target Actin, alpha skeletal muscle (gene ACTA1; UniProt P68133, human)
Clone AAOG-1
Host / Clonality / Isotype Rabbit / Monoclonal / Rabbit IgG

Validated Applications

Western blot 1:1000–1:5000
Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:50

Samples with a confirmed band (WB): HeLa cells, NIH/3T3 cells, C6 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human actin.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places actin, alpha skeletal muscle in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:5000; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 42.1 kDa; observed band at about 42 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human actin.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
