Product Overview

Clone DGI-3 is a rabbit monoclonal antibody that recognizes caspase-3 (gene CASP3) in human, mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 31.6 kDa; the supplier reports observed bands at 17,35 kDa.

Caspase-3 is the major executioner caspase of apoptosis. Activated by initiator caspases such as caspase-8 and caspase-9, it cleaves hundreds of cellular proteins, including PARP1, leading to the typical features of apoptotic cell death. Cleaved (active) caspase-3 is one of the most widely used apoptosis markers in Western blot and immunohistochemistry.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunoprecipitation 1:20

Samples with a confirmed band (WB): human HeLa cells, human 293T cells, human Jurkat cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Samples with confirmed staining (IHC): human melanoma tissue, human colon tissue, human lung large cells cancer tissue, human stomach tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human active + pro Caspase 3.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places caspase-3 in the cytoplasm. Tissue expression noted by UniProt (human): Highly expressed in lung, spleen, heart, liver and kidney. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on feline samples?
A.Reactivity with feline samples has not been tested by the supplier. Aligning the immunogen sequence with the feline ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 31.6 kDa; observed bands at about 17 and 35 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human active + pro Caspase 3.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
