Product Overview

Clone AOCB-1 is a rabbit monoclonal antibody that recognizes alpha-actinin-2 (gene ACTN2) in human, mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 103.9 kDa, and the supplier reports an observed band at about 104 kDa.

Alpha-actinin-2 (ACTN2) is the main alpha-actinin of cardiac and skeletal muscle. It cross-links actin filaments at the Z-disc and anchors the sarcomere, and variants in ACTN2 are associated with cardiomyopathies.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunoprecipitation 1:20

Samples with a confirmed band (WB): HepG2 cells, rat skeletal muscle tissue, rat heart tissue, mouse skeletal muscle tissue, mouse heart tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human ACTN2.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places alpha-actinin-2 in the cytoplasm. Tissue expression noted by UniProt (human): Expressed in both skeletal and cardiac muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
