Product Overview
Clone 21A54 is a rabbit monoclonal antibody that recognizes alpha-actinin-3 (gene ACTN3) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 103.2 kDa, and the supplier reports an observed band at about 103 kDa.
Alpha-actinin-3 (ACTN3) is an actin cross-linking protein found at the Z-discs of fast-twitch skeletal muscle fibers. A common nonsense variant (R577X) leaves many people without ACTN3, which has made it a well-known gene in studies of muscle performance.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|
Samples with a confirmed band (WB): mouse skeletal muscle.
Recommended loading (WB): 20–40 µg of total protein per lane.
Immunogen
Synthetic peptide based on the sequence of human ACTN3.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Expression restricted to fast (type 2) skeletal muscle fibers (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
