Product Overview
This rabbit polyclonal antibody detects adenosine deaminase (gene Ada) in mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 39.9 kDa, and the supplier reports an observed band at about 41 kDa.
Adenosine deaminase (ADA) converts adenosine and deoxyadenosine to inosine and deoxyinosine, controlling adenosine levels inside and outside cells. It is essential for lymphocyte development, and ADA deficiency causes a form of severe combined immunodeficiency (SCID).

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
| ELISA (capture antibody) | 1–5 µg/mL |
Samples with a confirmed band (WB): rat brain tissue, rat liver tissue, mouse liver tissue, mouse spleen tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat spleen tissue, mouse intestine tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Recombinant rat ADA fragment (Ala2–His241), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places adenosine deaminase in the cell membrane, cell junction and cytoplasmic vesicle lumen. Tissue expression noted by UniProt (rat): Detected in brain and liver (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
