Product Overview
Clone AAIC-1 is a rabbit monoclonal antibody that recognizes disintegrin and metalloproteinase domain-containing protein 10 (gene ADAM10) in human, mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 84.1 kDa, and the supplier reports an observed band at about 84 kDa.
ADAM10 is a membrane-anchored metalloprotease that cleaves the extracellular part of many surface proteins, a process called shedding. Its substrates include Notch, E-cadherin and the amyloid precursor protein, for which it is the main alpha-secretase, so ADAM10 is studied in development, neurodegeneration and cancer.

| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
| Immunoprecipitation | 1:20 |
Samples with a confirmed band (WB): human A431 cells, human Hacat cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human rectum adenocarcinoma tissue, human glioblastoma tissue, human ovarian serous adenocarcinoma tissue, human lung adenocarcinoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human ADAM10.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places disintegrin and metalloproteinase domain-containing protein 10 in the cell membrane, Golgi apparatus membrane, cell projection and cell junction. Tissue expression noted by UniProt (human): Expressed in the brain (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
