Product Overview
Clone AFCB-1 is a rabbit monoclonal antibody that recognizes double-stranded RNA-specific adenosine deaminase (gene ADAR) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 136.1 kDa, and the supplier reports an observed band at about 150 kDa.
ADAR1 (ADAR) is an RNA-editing enzyme that converts adenosine to inosine in double-stranded RNA. By editing the cell's own RNAs it prevents them from being sensed as viral, damping type I interferon responses, and its loss causes Aicardi-Goutières syndrome.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human Raji cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human ADAR1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Ubiquitously expressed, highest levels were found in brain and lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
