Product Overview

This rabbit polyclonal antibody recognizes adiponectin (gene ADIPOQ) in human, mouse and rat samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 26.4 kDa, and the supplier reports an observed band at about 160 kDa.

Adiponectin is a hormone secreted by adipocytes and one of the most abundant adipokines in plasma. Acting through the receptors ADIPOR1 and ADIPOR2, it activates AMPK and increases insulin sensitivity and fatty acid oxidation, and low circulating levels are associated with obesity and type 2 diabetes.

Observed MW 160 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

ELISA 0.1–0.5 µg/mL
IHC (frozen sections) 0.5–1 µg/mL (Rat, Human, Mouse)
IHC (paraffin sections) 0.5–1 µg/mL (Human, Mouse, Rat)
Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse)

Samples with confirmed staining (IHC): rat kidney tissue, mouse kidney tissue, human placenta tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Samples with confirmed staining (IHC): rat liver tissue.

Immunogen

Synthetic peptide from the C-terminal region of human ADIPOQ. It differs from the mouse and rat sequences at one residue.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt describes adiponectin as a secreted protein. Tissue expression noted by UniProt (human): Synthesized exclusively by adipocytes and secreted into plasma. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse); IHC (paraffin sections) 0.5–1 µg/mL (Human, Mouse, Rat); IHC (frozen sections) 0.5–1 µg/mL (Rat, Human, Mouse); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on primate samples?
A.Reactivity with primate samples has not been tested by the supplier. Aligning the immunogen sequence with the primate ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 26.4 kDa; observed band at about 160 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
