Product Overview

This rabbit polyclonal antibody detects beta-1 adrenergic receptor (gene ADRB1) in mouse and rat samples and is validated for Western blot. The predicted molecular weight is 51.3 kDa, and the supplier reports an observed band at about 51 kDa.

Beta-1 adrenergic receptor (ADRB1) is a Gs-coupled receptor for norepinephrine and epinephrine that raises cAMP. It is the main adrenergic receptor of the heart, increasing heart rate and contractility, and it is the target of beta-blocker drugs.

Target Beta-1 adrenergic receptor (gene ADRB1; UniProt P08588, human)

Validated Applications

Western blot 0.25–0.5 µg/mL (Mouse, Rat)

Samples with a confirmed band (WB): rat heart tissue, rat NRK cells, mouse heart tissue, mouse NIH/3T3 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide from the C-terminal region of human ADRB1. Sequence identity with the mouse and rat orthologs: 81.2%.

Reactivity Notes

The supplier lists reactivity with mouse and rat. UniProt places beta-1 adrenergic receptor in the cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 51.3 kDa; observed band at about 51 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human ADRB1. Sequence identity with the mouse and rat orthologs: 81.2%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
