Product Overview
This rabbit polyclonal antibody detects TLE family member 5 (gene TLE5) in human samples and is validated for ELISA, flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 22.0 kDa, and the supplier reports an observed band at about 22 kDa.
TLE5 (also called AES or Grg5) is a short member of the Groucho/TLE corepressor family that lacks the C-terminal WD40 domain. It can act as a dominant regulator of other TLE proteins and inhibits NF-κB-driven transcription. TLE5 is studied in Wnt and Notch signaling, differentiation and cancer.

Samples with a confirmed band (WB): human U-87MG cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human placenta tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): U87 cells.

Reactivity Notes
The supplier lists reactivity with human. UniProt places TLE family member 5 in the nucleus. Tissue expression noted by UniProt (human): Found predominantly in muscle, heart and Placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
