Product Overview
Clone GCI-1 is a rabbit monoclonal antibody that recognizes anterior gradient protein 2 homolog (gene AGR2) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 20.0 kDa, and the supplier reports an observed band at about 17 kDa.
Anterior gradient 2 (AGR2) is a protein disulfide isomerase family member located in the endoplasmic reticulum. It is needed for proper production and secretion of the mucin MUC2 by intestinal cells, and its frequent overexpression in adenocarcinomas has made it a subject of research on cell migration, growth and tumor biology.

Samples with a confirmed band (WB): human K562 cells, human RT4 cells, human Caco-2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

The supplier lists reactivity with human. UniProt describes anterior gradient protein 2 homolog as secreted and also located in the endoplasmic reticulum. Tissue expression noted by UniProt (human): Expressed strongly in trachea, lung, stomach, colon, prostate and small intestine. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
