Product Overview

This rabbit polyclonal antibody targets aryl hydrocarbon receptor repressor (gene AHRR) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 76.3 kDa, and the supplier reports an observed band at about 100 kDa.

The aryl hydrocarbon receptor repressor (AHRR) is a bHLH-PAS protein that dampens AHR signaling. It competes with AHR for partnering with ARNT and binds xenobiotic response elements, repressing genes such as CYP1A1 and forming a negative feedback loop in the response to dioxin-like compounds.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human)
IHC (paraffin sections) 2–5 µg/mL (Human)
Immunocytochemistry / Immunofluorescence 5 µg/mL (Human)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)

Samples with a confirmed band (WB): human THP-1 cells, human HeLa cells, human SiHa cells, human HEL cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human bladder urothelial carcinoma tissue, human colorectal adenocarcinoma tissue, human glioma tissue, human liver cancer tissue, human lung cancer tissue, human placenta tissue, human spleen tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with confirmed staining (ICC/IF): HeLa cells.

Samples with a confirmed signal (flow cytometry): HEL cells.

Immunogen

Recombinant human AHRR fragment (Lys410–Asp665), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human. UniProt places aryl hydrocarbon receptor repressor in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Highly expressed in testis, lung, ovary, spleen and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human); IHC (paraffin sections) 2–5 µg/mL (Human); Immunocytochemistry / Immunofluorescence 5 µg/mL (Human); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 76.3 kDa; observed band at about 100 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human AHRR fragment (Lys410–Asp665), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
