Product Overview
This rabbit polyclonal antibody detects interferon-inducible protein AIM2 in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 39.0 kDa, and the supplier reports an observed band at about 45 kDa.
AIM2 (absent in melanoma 2) is an interferon-inducible sensor that detects double-stranded DNA in the cytosol. On binding DNA it assembles an inflammasome with ASC (PYCARD) and caspase-1, leading to IL-1β and IL-18 maturation and pyroptosis, so it is studied in antiviral and antibacterial immunity and autoinflammation.

Validated Applications
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
|---|---|
| Western blot | 0.1–0.5 µg/mL (Human, Mouse) |
Samples with a confirmed band (WB): human A549 cells, human Daudi cells, human K562 cells, human HepG2 cells, mouse testis tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse kidney tissue, rat intestine tissue, human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human AIM2 fragment (Leu14–His215), expressed in E. coli. Sequence identity with the mouse ortholog: 53.8%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places interferon-inducible protein AIM2 in the cytoplasm, inflammasome and nucleus. Tissue expression noted by UniProt (human): Expressed in spleen, small intestine, peripheral blood leukocytes, and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
