Key elements and design rationale

  • Target: ALDOA (cofilin 2); UniProt: P04075
  • Antibody format: Mouse, Monoclonal, clone Clone: 6H8, Mouse IgG2b
  • Molecular weight: 39 kDa
  • Applications: WB, IHC, IF, ICC, Flow Cytometry, ELISA

Vendor description (summary): Boster Bio Anti-Aldolase/ALDOA Antibody Picoband® (monoclonal, 6H8) catalog # M05022-2.

Biological background

Biological context: Controls reversibly actin polymerization and depolymerization in a pH-sensitive manner. Its F-actin depolymerization activity is regulated by association with CSPR3. It has the ability to bind G- and F-actin in a 1:1 ratio of cofilin to actin. It is the major component of intranuclear and cytoplasmic actin rods. Required for muscle maintenance. May play a role during the exchange of alpha-actin forms during the early postnatal remodeling of the sarcomere.

Expression and localization notes: cellular localization: Cytoskeleton. Nucleus matrix., tissue context: Isoform CFL2b is expressed predominantly in skeletal muscle and heart. Isoform CFL2a is expressed in various tissues..

Common research applications

  • Western blotting (WB): Compare ALDOA levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of ALDOA in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify ALDOA-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: Aldolase A (ALDOA, or ALDA), also known as fructose-bisphosphate aldolase, is an enzyme that in humans is encoded by the ALDOA gene on chromosome 16. This gene encodes a member of the class I fructose-bisphosphate aldolase protein family. The encoded protein is a glycolytic enzyme that catalyzes the reversible conversion of fructose-1,6-bisphosphate to glyceraldehyde 3-phosphate and dihydroxyacetone phosphate. Three aldolase isozymes (A, B, and C), encoded by three different genes, are differentially expressed during development. Mutations in this gene have been associated with Glycogen Storage Disease XII, an autosomal recessive disorder associated with hemolytic anemia. Disruption of this gene also plays a role in the progression of multiple types of cancers. Related pseudogenes have been identified on chromosomes 3 and 10.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cytoskeleton. Nucleus matrix.
  • Tissue details: Isoform CFL2b is expressed predominantly in skeletal muscle and heart. Isoform CFL2a is expressed in various tissues.
  • Research category: Cell Adhesion Proteins,Epigenetics and Nuclear Signaling,Integrins,Mediator Complex,Neuroscience,Receptors,Transcription
