Product Overview
This rabbit polyclonal antibody recognizes alkaline phosphatase, tissue-nonspecific isozyme (gene ALPL) in human, mouse and rat samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 57.3 kDa, and the supplier reports an observed band at about 80 kDa.
Tissue-nonspecific alkaline phosphatase (TNAP, ALPL) is expressed in bone, liver and kidney. By hydrolyzing inorganic pyrophosphate, an inhibitor of mineralization, and pyridoxal 5′-phosphate, it is essential for bone and tooth mineralization, and ALPL mutations cause hypophosphatasia.

Samples with a confirmed band (WB): human HeLa cells, human HEK293 cells, rat kidney tissue, rat RH-35 cells, mouse kidney tissue, mouse HEPA1–6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:100, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): A431 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places alkaline phosphatase, tissue-nonspecific isozyme in the cell membrane, extracellular vesicle membrane, mitochondrion membrane and mitochondrion intermembrane space. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
