Vendor description (summary): Boster Bio Anti-alpha Adaptin/AP2A1 Antibody Picoband® catalog # A08806-1.

Biological background

Biological context: Protein phosphatase that inactivates multifunctional CaM kinases such as CAMK4 and CAMK2. Dephosphorylates and inactivates PAK. May play a role in the inhibition of actin fiber stress breakdown and in morphological changes driven by TNK2/CDC42. Dephosphorylates PRKAA2.

Expression and localization notes: cellular localization: Nucleus. Cytoplasm., tissue context: Expressed in fetal and adult brain. Also detected in fetal liver and skeletal muscle, but not in their adult counterparts..

Common research applications

  • Western blotting (WB): Compare AP2A1 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of AP2A1 in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify AP2A1-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: AP-2 complex subunit alpha-1 is a protein that in humans is encoded by the AP2A1 gene. This gene encodes the alpha 1 adaptin subunit of the adaptor protein 2 (AP-2) complex found in clathrin coated vesicles. The AP-2 complex is a heterotetramer consisting of two large adaptins (alpha or beta), a medium adaptin (mu), and a small adaptin (sigma). The complex is part of the protein coat on the cytoplasmic face of coated vesicles which links clathrin to receptors in vesicles. Alternative splicing of this gene results in two transcript variants encoding two different isoforms. A third transcript variant has been described, but its full length nature has not been determined.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Nucleus. Cytoplasm.
  • Tissue details: Expressed in fetal and adult brain. Also detected in fetal liver and skeletal muscle, but not in their adult counterparts.
  • Research category: Growth Factors/Hormones,Signal Transduction
