Product Overview
Clone 22A24 is a rabbit monoclonal antibody that recognizes THO complex subunit 4 (gene ALYREF) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 26.9 kDa, and the supplier reports an observed band at about 32 kDa.
ALYREF (also called ALY or THOC4) is an mRNA export adaptor in the TREX complex. It associates with spliced, capped mRNAs and hands them to the export receptor NXF1, coupling mRNA processing to nuclear export, and it has also been described as a reader of 5-methylcytosine on mRNA.

Samples with a confirmed band (WB): human 293T cells, human SH-SY5Y cells, human U251 cells, human HepG2 cells, rat RH35 cells, rat C6 cells, mouse HEPA1–6 cells, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human lymphoma tissue, human glioma tissue, human testis cancer tissue, human bladder cancer tissue, human placenta tissue, human renal cancer tissue, human cervical cancer tissue, human breast cancer tissue, human thyroid cancer tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places THO complex subunit 4 in the nucleus, nucleus speckle and cytoplasm. Tissue expression noted by UniProt (human): Expressed in a wide variety of cancer types. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
