Product Overview
This rabbit polyclonal antibody targets E3 ubiquitin-protein ligase AMFR in human, mouse and rat samples and is validated for flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 73.0 kDa, and the supplier reports an observed band at about 73 kDa.
AMFR (gp78) is an ER membrane E3 ubiquitin ligase that works with VCP/p97 in ER-associated degradation (ERAD). It tags misfolded or regulated ER proteins, such as sterol-bound HMG-CoA reductase and mutant CFTR, for proteasomal degradation, and it is studied in protein quality control and cholesterol regulation.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Rat) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human Daudi cells, rat kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human placenta tissue, rat kidney tissue, mouse intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): T47D cells.
Samples with a confirmed signal (flow cytometry): SiHa cells, U87 cells.
Immunogen
Recombinant human AMFR fragment (Glu553–Ser643), expressed in E. coli. Sequence identity with the mouse ortholog: 89%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places E3 ubiquitin-protein ligase AMFR in the endoplasmic reticulum membrane. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
