Product Overview
Clone CDD-16 is a rabbit monoclonal antibody that recognizes 5'-AMP-activated protein kinase catalytic subunit alpha-1 (gene PRKAA1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 64.0 kDa, and the supplier reports an observed band at about 64 kDa.
AMPK α1 (PRKAA1) is one of the two catalytic subunits of AMP-activated protein kinase, the cell's main energy sensor. When ATP falls, AMPK phosphorylates enzymes such as acetyl-CoA carboxylase and HMG-CoA reductase to switch off lipid and protein synthesis and switch on energy-producing pathways, so it is central to metabolism and diabetes research.

Samples with a confirmed band (WB): human HeLa cells, human U-87MG cells, human PC-3 cells, human A549- WT cells, human A549-PRKAA1 KO cells, rat PC-12 cells, rat RH35 cells, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung adenocarcinoma tissue, human tonsil tissue, human pituitary tumor tissue, human kidney tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human AMPK alpha 1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places 5'-AMP-activated protein kinase catalytic subunit alpha-1 in the cytoplasm, nucleus and late endosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
