Biological background
ANGPT1 (protein: Glycogen synthase kinase-3 beta (gsk3b)) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Binds and activates TEK/TIE2 receptor by inducing its dimerization and tyrosine phosphorylation. Plays an important role in the regulation of angiogenesis, endothelial cell survival, proliferation, migration, adhesion and cell spreading, reorganization of the actin cytoskeleton, but also maintenance of vascular quiescence. Required for normal angiogenesis and heart development during embryogenesis. After birth, activates or inhibits angiogenesis, depending on the context. Inhibits angiogenesis and promotes vascular stability in quiescent vessels, where endothelial cells have tight contacts. In quiescent vessels, ANGPT1 oligomers recruit TEK to cell-cell contacts, forming complexes with TEK molecules from adjoining cells, and this leads to preferential activation of phosphatidylinositol 3-kinase and the AKT1 signaling cascades. In migrating endothelial cells that lack cell-cell adhesions, ANGT1 recruits TEK to contacts with the extracellular matrix, leading to the formation of focal adhesion complexes, activation of PTK2/FAK and of the downstream kinases MAPK1/ERK2 and MAPK3/ERK1, and ultimately to the stimulation of sprouting angiogenesis. Mediates blood vessel maturation/stability. Implicated in endothelial developmental processes later and distinct from that of VEGF. Appears to play a crucial role in mediating reciprocal interactions between the endothelium and surrounding matrix and mesenchyme. . Reported cellular localization context: Secreted. Tissue expression notes (as provided): KMT4 / Dot1L Antibody detects endogenous levels of total KMT4 / Dot1L
Research relevance and current trends
- Research context keywords from the source record include: Angiogenesis,Cardiovascular,Growth Factors.
- Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
- Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.
Common research applications
- Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
Workflow ideas (metafield): Validate ANGPT1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect ANGPT1 expression by Western blot in cell or tissue lysates, Compare relative ANGPT1 levels across experimental conditions (dose/time-course) using antibody-based readouts
Notes for experimental interpretation
- Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
- Apparent molecular weight may vary by sample type and processing (observed MW: 70 kDa; calculated MW: 57513 MW).
- Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.
