Product Overview

Clone 23T77 is a rabbit monoclonal antibody that recognizes transcription factor AP-2-alpha (gene TFAP2A) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 48.1 kDa, and the supplier reports an observed band at about 48 kDa.

AP-2α (TFAP2A) is a transcription factor that binds GC-rich AP-2 sites as a dimer. It is particularly important in neural crest cells and for development of the face, eye, limbs and body wall, and TFAP2A mutations cause branchio-oculo-facial syndrome.

Immunoprecipitation 1:50
Flow cytometry 1:50

Samples with a confirmed band (WB): human SIHA cells, human A431 cells, human 293T cells, rat C6 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human AP2 alpha.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places transcription factor AP-2-alpha in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
