| Immunogen | A synthesized peptide derived from human APC Tumor suppressor. Promotes rapid degradation of CTNNB1 and participates in Wnt signaling as a negative regulator. APC activity is correlated with its phosphorylation state. Activates the GEF activity of SPATA13 and ARHGEF4. Plays a role in hepatocyte growth factor (HGF) -induced cell migration. Required for MMP9 up-regulation via the JNK signaling pathway in colorectal tumor cells. Acts as a mediator of ERBB2-dependent stabilization of microtubules at the cell cortex. |

Product Overview
Clone FAD-1 is a rabbit monoclonal antibody that recognizes adenomatous polyposis coli protein (gene APC) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 311.6 kDa, and the supplier reports an observed band at about 160 kDa.
Adenomatous polyposis coli (APC) is a tumor suppressor and core part of the β-catenin destruction complex, which keeps Wnt signaling off by targeting β-catenin for degradation. APC also binds microtubules and actin to regulate cell migration, and its mutation is the initiating event in most colorectal cancers and in familial adenomatous polyposis.

Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human APC Tumor suppressor.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places adenomatous polyposis coli protein in the cell junction, cytoplasm, cell projection and cell membrane. Tissue expression noted by UniProt (human): Expressed in a variety of tissues: brain, small intestine, colon, thymus, skeletal muscle, heart, prostate, lung, spleen, ovary, testis kidney, placenta, blood and liver. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
