Product Overview
This rabbit polyclonal antibody detects autophagy protein 5 (gene ATG5) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 32.4 kDa, and the supplier reports an observed band at about 50–52 kDa.
ATG5 is a core autophagy protein that is covalently linked to ATG12 by the enzymes ATG7 and ATG10. The ATG12-ATG5 conjugate, bound to ATG16L1, drives LC3 lipidation and autophagosome formation, and ATG5 knockout cells and animals are standard tools for studying autophagy.

Samples with a confirmed band (WB): human A549 cells, human HeLa cells, human HepG2 cells, rat C6 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human ovarian cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Synthetic peptide from the middle region of human APG5L. The peptide sequence is conserved in mouse. It differs from the rat sequence at one residue.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places autophagy protein 5 in the cytoplasm and preautophagosomal structure membrane. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
