Product Overview

Clone ACFH-12 is a rabbit monoclonal antibody that recognizes low-density lipoprotein receptor-related protein 8 (gene LRP8) in human, mouse and rat samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 105.6 kDa, and the supplier reports an observed band at about 106 kDa.

LRP8, also called ApoER2, is a member of the LDL receptor family that binds Reelin and apolipoprotein E-containing ligands. Together with the VLDL receptor it relays Reelin signals to the adaptor DAB1, guiding neuronal layering in the developing forebrain, and it also contributes to sperm maturation in the epididymis and to selenium uptake by selenoprotein P.

Validated Applications

Western blot 1:500–1:2000
Immunoprecipitation 1:20

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human apoER2.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt describes low-density lipoprotein receptor-related protein 8 as secreted and also located in the cell membrane. Tissue expression noted by UniProt (human): Expressed mainly in brain and placenta.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
