Product Overview
This rabbit polyclonal antibody targets adenine phosphoribosyltransferase (gene Aprt) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 19.7 kDa, and the supplier reports an observed band at about 20 kDa.
Adenine phosphoribosyltransferase (APRT) is a purine salvage enzyme that converts free adenine and PRPP into AMP. This recycling route is less energy-costly than making purines from scratch, and loss of APRT activity leads to the buildup of poorly soluble 2,8-dihydroxyadenine, which can form kidney stones.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
Samples with a confirmed band (WB): human MCF-7 cells, human 293T cells, human SiHa cells, rat liver tissue, rat kidney tissue, mouse liver tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human intestinal cancer tissue, human mammary cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Synthetic peptide from the C-terminal region of mouse APRT. Sequence identity with the human and rat orthologs: 86.4% and 95.5%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places adenine phosphoribosyltransferase in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
