Product Overview

Clone 4C10 is a mouse monoclonal antibody that recognizes arylsulfatase A (gene ARSA) in human, mouse and rat samples and is validated for flow cytometry, IHC and Western blot. The predicted molecular weight is 53.6 kDa, and the supplier reports an observed band at about 54 kDa.

Arylsulfatase A (ARSA) is a lysosomal enzyme that removes the sulfate group from sulfatides (cerebroside-3-sulfate), key lipids of myelin. Deficiency of ARSA causes metachromatic leukodystrophy, in which sulfatides accumulate and damage the nervous system.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)
IHC (paraffin sections) 0.5–1 µg/mL (Human)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human, Mouse)

Samples with a confirmed band (WB): rat testis tissue, rat liver tissue, rat brain tissue, rat lung tissue, mouse testis tissue, mouse liver tissue, mouse brain tissue, mouse lung tissue, mouse HEPA1–6 cells, human A375 cells, human A549 cells, human SMMC-7721 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): human lung cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Samples with a confirmed signal (flow cytometry): ANA-1 cells, Raji cells.

Immunogen

Synthetic peptide from the C-terminal region of human ARSA. It differs from the mouse sequence at six residues.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places arylsulfatase A in the endoplasmic reticulum and lysosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 0.5–1 µg/mL (Human); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human, Mouse). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 53.6 kDa; observed band at about 54 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human ARSA. It differs from the mouse sequence at six residues.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
